Abstract

Processing of cryo-EM data to execute structural analyses of aquaporins. Aquaporins (AQPs) are membrane proteins facilitating the flux of water and/or small neutral solutes such as glycerol over cell membranes. Drugs targeting aquaporins have broad potential clinical applications, including cancer, obesity, skin diseases among others. Here, we aim to process the first structure of an aquaporin binding with a drug-like small molecule. The structure in combination with the functional analyses will serve as a framework for the development of AQP inhibitors. As AQP deletion has been shown to be beneficial in different types of cancers as well as other diseases, the inhibition mechanism will provide a molecular basis for new therapeutic strategies in cancer.
